He was an advocate of Indian removal from the eastern United States, proposing an Indian state in what is now Kansas, Nebraska, and Oklahoma so Native Americans could be free from the "corrupting influences attending association with the frontier people of that early period." Isaac McCoy was a key part in the work of the Board of Foreign Missions in America and the Committee on Indian Affairs on the issues of Native American emigration and the Indian Removal Act.

This manuscript lists the names of American Indian people who were identified as having partial Omaha ancestry and who were born before 1830. This document is part of the Isaac McCoy collection. A Christian missionary, McCoy lobbied Washington officials in the 1820s for the removal of Indian tribes east of the Mississippi to unoccupied areas in the West. By the 1830s McCoy worked for the United States government, selecting and surveying locations for the immigrant Indians and establishing and maintaining missions and schools.

An entry on September 2, 1838, acknowledges the receipt of a letter informing of the death of William Clark, who had served as Superintendent of Indian Affairs in the central district until his death.
